MIVO® Gut-on-Chip: Humanized Intestinal Permeability Assay
This application note presents the MIVO® gut-on-chip platform combined with Caco-2/HT-29 co-cultures or 3D intestinal tissues to evaluate permeability of reference sugars (dextran, lactulose, mannitol). Results demonstrate in vivo-like absorption profiles and clinical data correlation, positioning MIVO® as a reliable, humanized 3R-compliant tool for preclinical ADME and formulation screening.
Intestinal permeability is a key factor in regulating the absorption of drugs and other compounds. The MIVO® gut-on-chip system was used in combination with either Caco-2/HT-29 cellular models or 3D-reconstructed tissues to assess the absorption of non-metabolized reference sugars, specifically dextran, lactulose and mannitol, to demonstrate the reliability of the model.
Sugar absorption profiles reached an in vivo-like plateau, confirming the physiological relevance of the tested models. Moreover, both the percentage of dextran absorbed and the lactulose-mannitol ratio within the MIVO® gut-on-chip were in line with clinical data.
Therefore, the combination of MIVO® with innovative intestinal models represents a reliable and predictive in vitro platform for carrying out intestinal absorption assays using a fully humanized 3R approach.
