The oral mucosa has a thin epithelium and a rich vascularization that facilitate the absorption of xenobiotics, whose chemical-physical properties influence their permeation, following oral administration. The assessment of the permeability of a substance and the estimation of its absorption help to identify potential candidates, such as drugs or bioactive functional compounds with nutritional or physiological effect.

Today, the number of in vitro assays able to mimic the real physiological conditions present in vivo remains limited and therefore the challenge is to develop advanced in vitro models, for which the key parameters such as permeability of substances through membrane barriers and their integrity, can be easily controlled and monitored with a reduction in experimental times and costs.

In this work we propose a new in vitro device (MIVO®) that allows to cultivate an oral epithelial tissue model under physiological flow conditions, in order to better predict the response in vivo, for an absorption study of S-Adenosyl-L- Methionine (SAM-e), a donor molecule of methyl groups in the synthesis of neurotransmitters, contained in an innovative orobuccal formulation.
