To date, preclinical safety and efficacy assessments of immunotherapies are carried out through extensive in vitro cultures addressing the cellular and molecular basis of immune responses, in-vivo xenografts and genetically engineered animal models.
However, both approaches show limitations, and the success of immunotherapeutic approaches strictly depends on the development of highly reliable and predictive preclinical models.

Cells Recruitment

IMMUNE CELLS RECRUITMENT

Immune cells extravasation, as well as their phenotype, is assessed recovering the cells from the medium, both surrounding the 3D culture and in circulation and analysing them by flow cytometry. Specific infiltration is analysed by immunofluorescence and confocal microscopy

Cytokines Release

CYTOKINE RELEASE KINETIC

Cytokine release can be assessed over time by easily sampling media through the three-way valve. Specific cytokines quantification can be performed by ELISA or similar detection methods

Immune Response

INFLAMMATION & IMMUNE RESPONSE

Activation of immune cells is evaluated by recovering the cells from circulating medium and assessing them for their phenotype by flow cytometry
